If you cheated on him and denied it, it would absolutely destroy him because though he can pick up on it because of his deductions, he would try to put it from his mind. That will lead to serious self doubt. Then eventually he would catch you out, and all of his deductions will be proven right. It's one of the few circumstances that he desperately wants to be wrong.

If you were seriously hurting emotionally and he didn't pick up on it in time. He knows exactly how dangerous our minds can be, and he takes mental heath very seriously. But because he is out with Sherlock so often, he's not always there to see the signs or comfort you. If his absence caused you to hurt yourself or worse, he would never forgive himself, and would spiral into his own depression.

If a rival criminal ever tried to get back at him through you. There are many circumstances this could lead to that would break him, but one of the worst would be if you were both captured and he had to watch you be tortured because of his rivalry. The images of you, in immense pain, would burn into his brain along with the sound of your agonised cries. It would most likely drive him insane.

If you ever got bored of him. He thinks he is a boring person, who lives a largely boring life. If Sherlock didn't exists, he doubts he would ever be able to solve the crimes that truly matter, and then he would lose his job which would not only make him more boring, but hopeless. If you were to get bored of him too, he would completely break.
MYCROFT

Losing you because of his difficulty to show his care, which he actually shows quite clearly towards Sherlock. There are only two things Mycroft truly and deeply cares about: his family and you. A lot of his time is spent trying to protect Sherlock and keeping him under control. That leaves the potential for you to feel lonely and less loved, which of course isn't at all the case. He doesn't want you to lose interest or feel unloved because he is better able to show his love for Sherlock than you - he tails Sherlock and checks up on him frequently, but knows he doesn't have to do that with you, but he struggles to find other ways to show his love.
